Project Description: 


Variants in genes encoding chromatin modifying and interacting proteins are frequent causes of neurodevelopmental disorders. This PhD project will focus on KDM5B, which encodes a histone lysine demethylase. KDM5B loss of function variants are associated with intellectual disability, autism spectrum disorders and cognitive ability in the general population. Using a combination of gene targeting and acute knock-down approaches, we have defined autism-like and learning and memory phenotypes in KDM5B-deficient mouse models.


The over-all aim of this project is to perform an in-depth analysis of the cellular and molecular mechanisms affected by KDM5B deficiency in the context of these behavioural phenotypes and to test whether targeting these mechanisms can restore normal function.


Specific objectives:


  1. To use a combination of single cell genomics, immunostaining and conditional gene deletion approaches to identify the brain regions and cell types affected by KDM5B deficiency.

  2. To use acute and in vivo electrophysiological approaches to define synaptic and circuit abnormalities in KDM5B-deficient mouse models.

  3. To use genetic and pharmacological approaches to target and identify the mechanisms responsible for neurodevelopmental phenotypes that can be targeted for therapeutic benefit.      

This award provides annual funding to cover Home tuition fees and a tax-free stipend. For students who pay Home tuition fees the award will cover the tuition fees in full, plus at least £18,622 per year tax-free stipend.  Students who pay international tuition fees are eligible to apply, but should note that the award will only provide payment for part of the international tuition fee and no stipend.  


International applicants need to be aware that you will have to cover the cost of your student visa, healthcare surcharge and other costs of moving to the UK to do a PhD. 


The conditions for eligibility of home fees status are complex and you will need to seek advice if you have moved to or from the UK (or Republic of Ireland) within the past 3 years or have applied for settled status under the EU Settlement Scheme.
